Nucleation and growth of polymorphs of pharmaceuticals on chemically modified surfaces

Branko Zugic, Kasim Biyikli, Garrett Ebersole, Joshua Allor, and John C. MacDonald. Worcester Polytechnic Institute, Worcester, MA

We are investigating whether nucleation and growth of polymorphs of pharmaceuticals such as barbital can be controlled on surfaces using self-assembled monolayers (SAMs). We have crystallized these compounds from solutions placed on SAMs functionalized with a variety of polar and nonpolar organic groups. Growth of polymorphs has been achieved by slow evaporation of solvent at RT (thermodynamic conditions) and by rapid cooling of hot solutions to RT (kinetic conditions). Temperature was controlled using a heating/cooling stage built in our laboratory.
