Azaspiracid-1, a toxin found in European shellfish, was first observed in the mid 1990's when several individuals became ill from eating mussels harvested off the coast of Western Ireland. Additional excitement was created by the revelation that the originally proposed structure of azaspiracid-1 (1) had been mis-assigned. The genesis of the error was believed to be in the ABCDE northern portion of the molecule. Our contributions towards the structure revision of azaspiracid-1 (2) will be discussed, including the successful synthesis of C1-C26 northern portion of the revised azaspiracid structure 2. Additional progress towards the total synthesis of 2 will also be presented.
