Study of Mast Cell Activation by the Combination of Atomic Force Microscopy and Confocal Laser Scanning Microscopy

A combined confocal laser scanning microscopy (CLSM) and atomic force microscopy (AFM) is utilized to investigate the morphological changes of mast cells upon IgE cross-linking by dinitrophenyl-bovine serum albumin (DNP-BSA). The high-resolution AFM topography acquired in PBS reveal the 3D morphological changes of the mast cell membrane upon activation. Membrane ridges with height ranging from 1 to 2 ėm are characteristic features for activated cells. Rich structural information of these mast cells at various stages of growth and activation will also be presented.
