Ethyl 4-(2-methyl-5,5-dimethyl-1,3-dioxan-2-yl)-5-formylisoxazole-3-carboxylate, a Branched Pentacarbonyl Synthon

Isoxazoles have served as useful 1,3-carbonyl synthons, exemplified by the Stork annulation, and have been used as polyketide synthons by Harris. For a number of years we have applied isoxazolyl-4-oxazolines successfully in several applications as branched tricarbonyl equivalents. We now report the preparation of Ethyl 4-(2-methyl-5,5-dimethyl-1,3-dioxan-2-yl)-5-formylisoxazole-3-carboxylate, a branched pentacarbonyl synthon which has each carbonyl differentially functionalized, we will discuss its preparation, optimization, and outline several applications in progress in our laboratories.
